Changing root user password

I cannot for the life of me figure out how to change the root user's password.
I tried directly changing it through phpMyAdmin in the mysql database but neither setting it directly as the password or encrypting it with the many hashing functions there are seem to work.
Any ideas?

I followed your manual step by step and still get the following error:

#1045 - Access denied for user 'root'@'localhost' (using password: NO)
